Current report

We have initiated Phase-1 start up activities with AN2-502998 (formerly known as AN15368), an investigational, boron-based small molecule in development for the treatment of chronic Chagas disease. In October 2023, we announced an exclusive license agreement with the University of Georgia Research Foundation to advance the development of AN2-502998, originally discovered by researchers at Anacor, in close collaboration with the University of Georgia. AN2-502998 is the only compound of which we are aware to have demonstrated curative activity in preclinical studies across multiple species, including in non-human primates with long-term, naturally acquired chronic infections of diverse T. cruzi genetic types. We anticipate completion of the Phase 1 study in the second half of 2025.

Current report

The Company is pursuing a number of oncology targets where we believe boron chemistry offers a competitive advantage in terms of binding-site differentiation, pharmacodynamics, drug-like properties and IP, including initially ENPP1 and PI3Kα. The unique binding modes of boron-containing compounds enable the discovery of inhibitors with high ligand efficiency against targets considered undruggable or difficult to access with traditional chemistry approaches. Boron chemistry has produced first-in-class molecules against a number of targets including CPSF3 (AN2-502998 and acoziborole) and LeuRS (epetraborole, GSK656/AN10070 and tavaborole). The Company has discovered preclinical compounds with profiles that are sub-nanomolar, highly selective and have excellent oral pharmacokinetics. We anticipate advancing the first oncology compound(s) into development later this year with potential clinical proof of concept within the Company’s current cash runway.

Current report

We are studying epetraborole for the treatment of acute melioidosis. We completed enrollment in a 200-patient observational trial (non-epetraborole treatment) in October 2024 and expect to announce topline data in the second quarter of 2025. This data will inform a Phase 2 proof of concept study that is planned to initiate start up activities in the second half of 2025. Melioidosis is a deadly bacterial infection and global bioterrorism threat with a 90-day mortality rate approaching 40% using standard of care (SOC) drugs ceftazidime or meropenem. The aim of the program is to meaningfully lower the expected mortality rate by dosing epetraborole on top of SOC.

Current report

Finally, the Company continues to evaluate the potential of epetraborole in NTM, where we believe that clinical trials to date provide significant enabling data in M. abscessus. On May 1, 2025, we announced the truncated Phase 3 portion of the EBO-301 study did not meet its primary endpoint on improvement of Quality of Life – Bronchiectasis (QOL-B) respiratory domain patient reported outcome instrument (change from baseline to month 6). The study population included patients with severe, advanced MAC lung disease with a long duration of disease, high rates of cavitary/fibrocavitary disease, and multidrug resistance to standard of care antibiotics at baseline.

Previous report

Since launching operations in November 2019, we have devoted substantially all of our R&D resources to developing our initial product candidate, epetraborole, which we are developing for treatment-refractory Mycobacterium avium complex (“MAC”) lung disease, a form of NTM lung disease. On August 8, 2024, we announced topline results from the Phase 2 part of the EBO-301 Phase 2/3 study evaluating epetraborole on top of optimized background regimen (“OBR”) in treatment-refractory MAC lung disease. The Phase 2 part of the study met its primary objective of demonstrating the potential validation of a novel patient-reported outcome (PRO) tool and a higher PRO-based clinical response rate in the epetraborole + OBR arm (39.5%) vs. placebo + OBR (25.0%; treatment difference 13.9%, p=0.19). Sputum culture conversion at Month 6, a key secondary endpoint, was similar between treatment arms (13.2% in epetraborole + OBR vs. 10.0% placebo + OBR; treatment difference 3.4%, p=0.64). Epetraborole was generally well tolerated in the trial.

Previous report

After announcing these results, we commenced further review of the Phase 2 data to determine the best path forward for this program. To date, we have identified nominally statistically significant outcomes in two patient-reported-outcome (PRO) analyses that suggest clinical improvement in epetraborole patients over placebo patients. The first—a prespecified secondary endpoint of the Quality of Life (Bronchiectasis) (QoL-B) PRO—indicates that patients on epetraborole showed statistically significant symptom improvement when calculated using its least squares mean (LSM) method. Of note, this is the same endpoint Insmed, Inc. recently announced as the primary endpoint in its ENCORE Phase 3 trial for treatment-naïve MAC. The second—a post-hoc LSM calculation of the MACrO2 PRO results—also showed statistically significant clinical symptom improvement in epetraborole patients vs. placebo. We believe these two findings provide potential clinical proof of concept and that improvements in two PRO measures appear to align with FDA’s recommendation, reflected in a recent guidance document, that trials to support drug approvals in NTM utilize PRO-based clinical outcomes as the primary endpoint. We will continue our data analysis and plan to request an End-of-Phase-2 meeting with FDA in the first half of 2025 to discuss the potential for re-initiating a pivotal Phase 3 trial in TR-MAC.

Previous report

As we continue to evaluate future development opportunities for epetraborole in NTM, we have advanced our pipeline programs. In 2025, we anticipate initiating a Phase 1 trial in Chagas disease and a Phase 2 proof-of-concept trial in melioidosis. We also anticipate progressing our earlier-stage programs in oncology and infectious disease toward producing up to three development compounds in 2025.

Previous wording

We do not have any products approved for sale, and we have never generated any revenue from contracts with customers. We do not expect to generate any meaningful revenue unless and until we obtain regulatory approval for and commercialize any of our current and future product candidates and we do not know when, or if, those events will occur. Historically, we have incurred operating losses and negative cash flows as a result of ongoing efforts to develop our initial drug product candidate, epetraborole, including conducting ongoing preclinical and nonclinical studies, clinical trials, registration API and drug product materials manufacturing, and providing general and administrative support for these operations. We expect our negative cash flows to increase significantly over the next several years as we advance our product candidates through clinical development, seek regulatory approval, prepare for and, if approved, proceed to commercialization, and continue our research and development efforts. We are subject to all the risks typically related to the development of new product candidates, and we may encounter unforeseen expenses, difficulties, complications, delays, and other unknown factors that may adversely affect our business. Moreover, we expect to continue to incur costs associated with operating as a public company. We anticipate that we will need substantial additional funding in connection with our continuing operations, as we do not expect positive cash flows from operations in the foreseeable future.
