Dear Summit Therapeutics Inc. Stockholder:
You are cordially invited to attend a special meeting of stockholders (the “Special Meeting”) of Summit Therapeutics Inc. (the “Company,” “Summit,” “we,” “us” or “our”), which will be held on November 4, 2026 at 11:00 a.m. Eastern Time. The Special Meeting will be conducted virtually via live webcast. You will be able to vote and submit your questions during the meeting by visiting www.virtualshareholdermeeting.com/SMMT2026SM; to participate in the Special Meeting, you will need the control number included on your notice or proxy card. Please have your notice or proxy card in hand when you visit the website. During the Special Meeting, stockholders will be asked to vote on the following proposals, as more fully described in the accompanying proxy statement (“Proxy Statement”):

Our Board of Directors has fixed the close of business on October 1, 2026 as the record date for the Special Meeting. Only stockholders of record on October 1, 2026 are entitled to notice of and to vote at the Special Meeting. This Proxy Statement is being mailed to stockholders on or about October 19, 2026. It is important that your shares are represented and voted at the Special Meeting.
